1. If a guest desires no further coffee, the host should not insist.

2. If a guest doesn't desire any more coffee, the host should not insist.

Are both 1 and 2 acceptable to you?

Yes, but it sounds a bit stiff and formal. I would change 'desire' to 'want'.
